Restriction digests of the clone give the following sizes (kb): PstI--9.0, 7.2, 4.3 (doublet), 4.1, 3.5, 2.3, 2.0; BglII--9.3 (doublet), 6.0, 5.6, 3.7, 3.2, 2.0, 1.9; EcoRI--6.2, 5.6, 4.8, 3.5, 2.9 (doublet), 2.6 (doublet), 2.1, 2.0 (doublet), 1.7, 0.9; HindIII--16.0, 13.0, 9.0, 7.0; BamHI--23.3, 13.0, 2.2. The insert can be excised from the vector with EcoRI, resulting in a 5.5 kb EcoRI vector fragment. IMPORTANT: To prevent amplification of a rearranged and/or deleted cosmid, we recommend streaking on LB + amp plates at 30C and picking small colonies for liquid culture. The HincII polymorphisms have a combined PIC value of 0.81 and a combined heterozygosity of 83%. Enzyme(s) not detecting polymorphism: BamHI, EcoRI, PstI, RsaI (panel of 5 individuals). |
